MSLN

MSLN,即间皮素(Mesothelin),是一种糖基磷脂酰肌醇(GPI)锚定的细胞表面糖蛋白,由MSLN基因编码的前蛋白经过蛋白水解后产生巨核细胞增强因子和间皮素两种蛋白产物。
MSLN的作用机制涉及其在细胞表面的表达,通过与配体如CA125/MUC16的高亲和力相互作用促进异型细胞粘附,进而影响肿瘤的转移。此外,MSLN的过表达能够激活NF-κB、MAPK和PI3K-激酶促进细胞增殖和抵抗凋亡的细胞内通路。MSLN在正常组织中的表达有限,但在卵巢癌、乳腺癌、结直肠癌和胰腺癌等几种实体肿瘤中的过度表达,与较低的存活率相关,是肿瘤特异性治疗的理想靶点。
