FLT3
FLT3，即FMS样酪氨酸激酶3，别名CD135。它是一种III型受体酪氨酸激酶（RTK），在造血干细胞、前体B细胞等的增殖、分化以及存活中具有重要作用。FLT3基因突变是急性髓系白血病（AML）患者中最常见的基因改变之一，也是预后不良的重要因素。这些突变导致FLT3受体在不依赖FLT3配体（FL）的情况下，持续自身磷酸化，异常激活下游的信号通路，如PI3K/AKT、RAS/MAPK/ERK和STAT5等，从而促进细胞增殖和抑制凋亡。FLT3对于维持正常的造血功能至关重要。然而，当FLT3发生突变时，它可能导致白血病的发生。FLT3突变的AML患者通常具有较高的外周血白细胞计数、较差的临床预后，并且容易复发。FLT3抑制剂的开发为AML患者提供了新的治疗选择，这些抑制剂通过竞争性抑制FLT3受体中的ATP结合位点，导致细胞周期停滞和分化，从而抑制白血病细胞的增殖。
