C5

C5,即补体成分5,别名C5α、C5b等。它是人类补体系统的一部分,由肝脏细胞产生,是一种血清蛋白。C5的作用机制主要是通过补体级联反应被激活,C5可以被C4b2a(经典途径和凝集素途径)或C3bBb3b(旁路途径)裂解为C5a和C5b两个片段。C5a是一个具有强烈炎症活性的小分子肽,能够吸引和激活多种免疫细胞,如中性粒细胞和单核细胞,而C5b则启动膜攻击复合物(MAC)的形成,导致外来细胞的溶解。C5在生物学上的意义非常广泛,它在天然免疫和获得性免疫之间起着桥梁作用。C5的活化参与了清除病原体和受损细胞、调节炎症反应、促进免疫细胞的趋化和活化等重要过程。C5a-C5aR1信号通路在多种炎症性疾病中发挥作用,包括结直肠癌等肿瘤的发展,通过影响免疫微环境和细胞内信号传导来促进肿瘤的发生和发展。C5的异常活化与多种自身免疫性疾病和炎症性疾病的发生发展密切相关,因此,C5或其受体C5aR1已成为药物开发的潜在靶点 。

Then 10% normal goat serum was Incubated to block non-specific protein-protein interactions followed by the antibody (1µg/1*106cells) for 45 min at 4°C. The secondary antibody used was FITC-conjugated Goat Anti-Mouse IgG(H+L) at 1/200 dilution for 35 min at 4°C. Isotype control antibody (green line) was mouse IgG1 (1µg/1*106cells) used under the same conditions. Acquisition of >10, 021 events was performed.
