ANGPT2

血管生成素-2(ANGPT2),也常见为ANG-2。ANG-2基因定位于染色体的8p23,属于分泌型糖蛋白,分子量大约75kDa。研究表明,ANG-2是ANG-1的天然竞争拮抗剂,两者的受体都是Tie-2。ANG-1与Tie-2结合可激活PI3K/AKT信号通路,维持血管的完整和稳定,抑制肿瘤血管生成。阻滞ANG-1对肿瘤血管系统几乎没有影响,而阻滞ANGPT2却能造成肿瘤血管系统的损伤和Ang-1的活化,使肿瘤血管正常化,抑制肿瘤血管形成。因此,ANGPT2不仅参与了胚胎血管发育、血管重构等正常生理过程,且在肿瘤、创伤、炎症等多种病理过程中发挥了重要作用。目前,ANGPT2已成为免疫检查点和血管生成抑制剂的疗法靶点。
