    The DNA fragment corresponding to Trp27-Ser549 of the human NT5E antigen is fused with a 6xHis-tag at the C-terminus and then is expressed in the mammalian cells. The resulting product is the recombinant human NT5E protein. Its purity is over 95% measured by SDS-PAGE. On the gel, this protein was migrated to the molecular weight band of about 62 kDa. The endotoxin content of this NT5E protein is less than 1.0 EU/ug determined by the LAL method. Its bioactivity has been validated through an ELISA. In the functional ELISA, immobilized NT5E can bind to anti-NT5E rabbit monoclonal antibody, with the EC50 of 3.212-4.525 ng/ml. This NT5E protein is available now.

    NT5E, also called CD73, is a bifunctional GPI-anchored cell surface protein with both enzymatic and nonenzymatic functions in cells. As an ecto-5′-nucleotidase, CD73 catalyzes the hydrolysis of AMP into adenosine and phosphate. As a membrane receptor for extracellular matrix protein (ECM), CD73 also modulates cell interaction with ECM components such as laminin and fibronectin, to mediate the invasion and metastasis of tumor cells. Overexpression of CD73 has been detected in many cancers, including gastric cancer, breast cancer, and colorectal cancer, and has been associated the poor clinical outcomes.
